Advanced Training Program

Central Dogma is dedicated to provide in-depth practical knowledge and skill development opportunities  to graduate/postgraduate students and professionals in the field of biotechnology,  microbiology, zoology, biochemistry and related life sciences. The primary goal is to make the individual “Industry ready”.

These trainings are conducted one-on-one and hence give platform to the participant to gain expertise in various techniques. During the course of training, the participant can get repeated exposure to various techniques which help in developing expertise and gain confidence to function as an independent researcher. Additionally, it improves analytical capabilities and troubleshooting skills. The training is conducted by domain experts of Central Dogma who have several years of research experience in academia and industry

Eligibility:

  • Students in the final year of BE/BTech and ME/MTech/MSc who have industrial project as a part of the curriculum.
  • Students/Researchers/Professionals who have completed graduate and post graduate degrees and would like to build up the hands on skills and improve their employability

Advanced Techniques in Recombinant DNA Technology

 Duration: 15 Days

Time: 10am -2pm

Techniques Covered

  • Plasmid DNA (vector) isolation from bacterial culture using kit
    • Agarose gel electrophoresis
  • RFLP and Agarose gel electrophoresisDSC_0052
  • Restriction Digestion of Lambda Phage with enzymes
  • Polymerase Chain Reaction (PCR)
      • Theory and primer design
      • Gene amplification by PCR
  • Competent Dh5 alpha cells preparation and transformation of plasmid
  • Heatshock treatment, Plating the transformants on the LB agar +Amp plates and overnight incubation
  • Growth Curve of bacterial host strain used in Recombinant DNA technology
  • Casting of SDS-PAGE gels
  • Protein Sample loading on SDS-PAGE
  • Protein profile analysis by coomasie stain

 Gene cloning and Recombinant Selection

Duration: 21 days

Time: 10am -2pm

  • Expression Plasmid DNA (vector) isolation from bacterial culture using kit
    • Agarose gel electrophoresis
  • RFLP and Agarose gel electrophoresis
  • Digestion of Plasmid vector with specific restriction enzyme used for cloning
  • Purification of digested vector by gel extraction method using kit
    • Polymerase chain reaction of the target gene and purification
  • Polymerase Chain Reaction (PCR)
      • Theory and primer design
      • Gene amplification by PCR
  • Digestion of PCR product with specific restriction enzyme
    • Purification of digested PCR product using kit
    • Ligation of digested vector and Insert
  • Competent Dh5 alpha cells preparation and transformation of ligation mixture
  • Heatshock treatment, Plating the transformants on the LB agar +Amp plates and overnight incubation
  • Confirmation of recombinants by colony PCR
    • Agarose gel electrophoresis of colony PCR to confirm recombinants

Advanced techniques in Molecular Biology and Protein Expression

Duration: 4-5 weeks

Time: 5-6 hrs/day

Eligibility: Undergrad (Final Year)/Postgrad students/Completed master’s degree

  Techniques covered:

  • Plasmid DNA (vector) isolation from bacterial culture using kit
  • Agarose gel electrophoresis
  • Plasmid mapping using multiple restriction enzymes
  • Polymerase chain reaction of the target gene and purification
  • Digestion of PCR product with specific restriction enzyme
  • Purification of digested PCR product using kit
  • Digestion of cloning vector with specific restriction enzymes
  • Purification of digested vector by gel extraction
  • Ligation of digested PCR product and vector
  • Competent cells preparation of propagation host and transformation with ligation mix
  • Microbial growth media preparation/sterilization and plating
  • Selection of recombinants by colony PCR method
  • Agarose gel electrophoresis of colony PCR to confirm recombinants
  • Recombinant plasmid isolation
  • Competent cells preparation of expression host
  • Transformation of expression host with recombinant plasmid
  • Shake flask fermentation and induction for recombinant protein expression
  • Cell harvest and lysis for release of recombinant protein
  • Protein profile on SDS-PAGE and staining using coomassie blue and silver stain

Analytical Techniques in Food Technology

Duration: 3 weeks

Time: 5-6 hrs/day

Eligibility: Undergrad Final Year)/Postgrad students/Completed master’s degree

Microbial Techniques

  • Microbial count in contaminated food
  • Detection of Coliforms (Std and PCR based)
  • Studying the microbial contamination of food on different media
  • Isolation of bacteria, study of its colony characters and its identification upto genus level (will include performing various biochemical tests)
